Twilight is fluid, XHTML 1.0 Strict, and passed the Wave test. Cynthis Says it is WAI and US 508 compliant as far as can be tested by a computer.
The grey text on the blue background does not have enough contrast for regular reading or for the visually impaired. I put twilight on the home page for a couple of days to test across various screens and it was hard to read on every screen. If you plan to use Twilight, change the grey text to white.
The theme has HTML tables used for layout of non tabular data, an old idea from the days when Web browsers did not offer an alternative. You would only use Twilight if you had to support very old browsers.
